Beyond antibacterials – exploring bacteriophages as antivirulence agents

چکیده
Life-threatening infections caused by multidrug-resistant bacteria are becoming increasingly difficult to treat. There is growing interest in exploiting bacteriophages (or phages) combat bacterial infections. Phages often target surface structures that may also be important for virulence. Upon phage challenge, these molecules lost or modified, resulting resistance and possibly phenotypical conversion. Importantly, possible trade-offs include lower fitness, increased sensitivity antibiotics immune defense mechanisms, virulence attenuation. Although evolution of phage-resistance prevent, the trade-off phenomenon carries potential antibacterial therapy. Here we present some insights into molecular principles significance this coincidental interplay between phages, bacteria, cells, discuss prospect developing phage-derived products as antivirulence agents.

Bacteriophages: antibacterials with a future?
The hypothesis as to whether a benign species of bacteria could kill a virulent kind has to this point been untested. Recently it was shown that in the macrophage, bacteriophages, when properly introduced through a nonvirulent microbe, had a killing rate for virulent AIDS Mycobacterium tuberculosis and Mycobacterium avium far in excess of modern day antibiotics. The study in effect brought a na...

The continuing emergence of antibiotic-resistant pathogens is a concern to human health and highlights the urgent need for the development of alternative therapeutic strategies. Quorum sensing (QS) regulates virulence in many bacterial pathogens, and thus, is a promising target for antivirulence therapy which may inhibit virulence instead of cell growth and division. This means that there is li...

We read the editorial entitled “Non-antibiotic treatments for bacterial diseases in an era of progressive antibiotic resistance” by Steven M. Opal published in Critical Care [1]. We want to congratulate the author for this timely editorial and appreciate the efforts to highlight various non-antibiotic-based approaches being pursued for treating drug-resistant bacteria. We fully agree that there...

Antivirulence agents inhibit the production of disease-causing virulence factors but are neither bacteriostatic nor bactericidal. Antivirulence agents against methicillin-resistant Staphylococcus aureus (MRSA) strain USA300, the most widespread community-associated MRSA strain in the United States, were discovered by virtual screening against the response regulator AgrA, which acts as a transcr...

Hosts and their microbes have established a sophisticated communication system over many millennia. Within mammalian hosts, this dynamic cross-talk is essential for maintaining intestinal homeostasis. In a genetically susceptible host, dysbiosis of the gut microbiome and dysregulated immune responses are central to the development of inflammatory bowel disease (IBD). Previous surveys of stool f...
